Regulation of Puma by FBXL6

Abstract
This paper presents the regulation of PUMA by FBXL6 protein. PUMA (p53 upregulated modulator of apoptosis) is a Bcl-2 homology 3 (BH3)- only Bcl-2 family member and an important negotiator of p53-dependent and p53-independent cell death evoked by a large diversity of stimuli together with genotoxic stress. Here, we have performed SCF E3 ubiquitin ligase screen and known FBXL6 as a potential candidate macromolecule that prohibit the expression level of PUMA. We performed western blotting technique and concluded that FBXL6 regulate PUMA at the post translational levels but FBXL6 mediated PUMA regulation is lost after DNA damage in order to increase PUMA level to induce apoptosis.
